I have a 2006 E250, with the 4.6L. 235K miles.

I've changed the fuel filter numerous times in my ownership with no issues: purge pressure, unhook the battery, remove the filter, replace the filter, reconnect the battery, cycle to on 10 times, fire up.

This last time, however, I can't get it to fire up. I can hear the pump working. Starts fine with starter fluid, but won't start by itself. The only difference this time is that I was parked on an inclined driveway. I'm guessing there is air in the fuel line but uncertain on how to purge. Suggestions?

How much fuel in the tank?
More than one tank? If yes switch tanks

The design is self bleeding and a returnless system.
The fuel pump driver module on the frame rail determines pressure based on need. It varies voltage to the pump.
Hearing the pump and having enough pressure may be two different things.
Replacing the filter may be a coincidence.

Testing this requires a scanner/tester.

You could remove the new filter and see if there is fuel in it.

I got it to fire up. Took the filter off, and repeated the procedure. Started up, but threw lean mix codes. Drove it around and they went away. Maybe a lock in the line, or just an issue with the filter? It did have gas, but not as much as the old filter. One of those 40-minute jobs that turned into a two-hour mystery. But she's running!
